Nope. If you are using windows right click on the drive and select properties. The file system will probably be FAT for a flash device and FAT32 or NTFS for a hard disk.
Just did it with my (1GB) MP3 player - it claims to be FAT32. An older 64MB camera card admits to FAT (presumably 16?).
