I can't think of anything funny to say about it but maziresu suruto,
among the two choices I'd say "not" is more likely. Another
possibility is that it might be a translation from English or some
other language that attempts to render the joke straight into
Japanese. It's a standard joke in English but as far as i know, this
kind of humor is not exactly Japanese people's strongest suit. That's
my take on it.
Perhaps, I am saying the standard joke in English without knowing or
meaning everyday such as -- Many (such as Mammographers) are nice, but
some are not. I bet they thought I was joking. Oh, my god.
Moreover, I told this line to a Mammographer. What would she have
thought of it? Do you think she thought I was being ironical to her?
The joke is the deliberate use of a homophone with a silent letter in
place of the correct spelling (knot->not).
So, what is funny then? Don't say "Forget it". Explain until I
understand.
It's a visual pun.

When I was a kid, one of my brothers had a huge eraser, on which was
written, in large letters, "I never make BIG misteaks". "Misteak" is,
of course, a misspelling of "mistake", so the whole thing was ironic.

So, Paul's "Many words are pronounced according to the spelling but
some are not." is supposed to be "Many words are pronounced according
to the spelling but some are knot." and it's very funny?

I wouldn't say it's very funny, but it's good for a small chuckle or groan, depending about how you feel about puns.
