Evaluation of genetic divergence among Borrelia burgdorferi
isolates by use
of OspA, fla, HSP60, and HSP70 gene probes.

In order to assess the genetic variation of immunologically
relevant structures
among isolates of the Lyme disease spirochete, Borrelia burgdorferi,
three chromosomal
genes encoding flagellin (fla) and the heat shock proteins HSP60 and
HSP70, as well
as the plasmid gene encoding outer surface protein A (OspA), from 55
different European
and North American strains obtained from ticks and mammal hosts have
been investigated
by restriction fragment length polymorphisms (RFLPs). RFLPs of fla and
the HSP60
and HSP70 genes revealed two distinct banding patterns (A and B) for
each of the
three genes and allowed the definition of four genomic groups [AAA,
BBB, BBA, and
B(A/B)A] for the three chromosomal genes. On the other hand, RFLPs of
the OspA gene
revealed six distinct banding patterns (types I to VI) making up six
independent
genomic groups for the plasmid-encoded gene. Furthermore, we have
sequenced the
chromosomal HSP60 gene from B. burgdorferi ZS7 and the plasmid-encoded
OspA gene
from two strains, ZQ1 and 19857. Alignment of the deduced HSP60 amino
acid sequence
from B. burgdorferi ZS7 (genomic group AAA) to a previously published
HSP60 sequence
derived from strain ACA-1, which according to the proposed
classification is in
a different genomic group (BBA), revealed a sequence identity of >
99%. Similar
alignments of the OspA sequence of strain ZQ1 to those of other
isolates that were
published previously revealed sequence identities of between 70 and
94% among strains
of distinct OspA genomic groups. These data indicate the existence of
a restricted
number of species-specific subgroups and clearly show that genotypic
variation is
much more pronounced for the OspA gene than for fla and the HSP60 and
HSP70 genes.
A phylogenetic tree constructed on the basis of distance matrix
analyses of 12 OspA
sequences supports the proposed classification of genomic groups of B.
burgdorferi.

They could have used Borreliae-specific DNA of the non-variable non-
Osp, non-vmp,
non plsamid DNA or intragenic specific spacer RNA.
They all knew it, but never told anyone that looking for the OspA gene
was the same
thing as saying "only bipedal female Homo primates with 18 inch long
blonde
hair or longer may be diagnosed as 'women'"
We happen to be convinced the Lyme crooks went with a foreign company
(SmithKline
and Connaught, now Pastuer-Aventis) so as to be out of reach of a RICO
investigation.
Apparently that's what the USDOJ, Criminal Division thought, when they
referred

GlaxoSmithKline faces US scrutiny over Paxil suicide link

GlaxoSmithKline (GSK), the UK's largest drug maker, is facing new
scrutiny in
America in the wake of a British government inquiry that found the
company withheld
data on the suicide risk of an antidepressant.
A US senator today asked the food and drug administration (FDA), the
agency that
regulates American pharmaceuticals, to follow its UK counterpart in
probing whether
GSK concealed clinical trial evidence.
"If the company engaged in this behaviour in the UK, then I want to
make sure
that the same didn't happen here in the US," Grassley said.
"The FDA should investigate this question thoroughly and be
forthcoming about
its findings."
The drug in question - Paxil, marketed in the UK as Seroxat - has been
the subject
of suicide warnings since 2003, when the British government told
doctors not to
prescribe it to young adults. A warning to that effect was added to
its US labels
in 2006.
In responding to the UK conclusion that GSK failed to inform
authorities of Paxil's
suicide risk, Grassley quoted the Guardian's report on imminent
British legislation
requiring drug companies to promptly publicise clinical trial data.
The senator also pointed to a report recently unsealed by a US court
that found
GSK knew as early as 1989 that Paxil carried a heightened suicide risk
in adults
as well as children.
The report, by Harvard University psychiatrist Joseph Glenmullen, was
based on internal
GSK documents and intended for use in ongoing US court cases against
the company.
Glenmullen found that GSK inappropriately inflated the suicide risk of
a placebo
during clinical trials of the antidepressant, obscuring the increased
danger associated
with Paxil.
"It is my opinion, based on a reasonable degree of medical probability
… that
Paxil increases the risk of suicidality in adults," Glenmullen wrote.
"In
addition, GlaxoSmithKline was aware of this risk, but hid it."
GSK has challenged the Glenmullen report since it was first unsealed
in February
and defended its use of so-called "washout" data in analysing suicide
attempts made by patients taking Paxil during clinical trials.
Asked for comment on the senator's request for an FDA inquiry, a GSK
spokeswoman
pointed to comments the company made earlier this year responding to
Grassley's
concerns about the Glenmullen report.
"Dr Glenmullen has carefully selected excerpts from GlaxoSmithKline
documents
and made his own interpretations without looking at the totality of
the data,"
GSK said in February.
