If you have an inertial frame relative to which you can move them that way
No....you don't have to have an inertial frame.
If you want them to stay in sync you do. If not, then SR says your measurements will not be correct because the clocks will be out of sync. ( Refer to H-K experiment ) So any difference in OWLS would not be meaningful (unless you adjust the results for the non-inertial frame). So, it would be to your advantage to use a frame that is inertial (or close enough to it that the errors are minimal)
