George Takei was definately hired by 1965 when they made the second pilot for Star Trek after the first was rejected in late 1964. They filmed the second pilot in the summer of 1965.
I am not sure, but I think that Takei might have signed onto the original contract prior to the first pilot, but they were going to introduce his character later. (Similar to how they introduced Doctor McCoy in the series that was picked up). They had enough trouble with gender politics in the initial pilot that they dropped the interracial nature of the crew in the first pilot. That would be very close to that 1963 date.
Sulu predates everyone in the series except Spock and Kirk and was introduced in the same ep as Kirk.
