Perhaps a very advanced confocal microscope could begin to discern the
features of AP. However, it is IMO beyond the limits of LM.
Hmmm. For practical purposes you are right, but I'd say it's AT the
limits of LM really. I've "captured the dots" quite well on A.
Pellucida with a mercury lamp, bandpass soft-UV filter and (very) long
exposure on a cooled CCD. I used a 64/1.4 planapo objective and
Nomarski DIC to get a luminance channel and combined it with a lower
resolution brightfield shot to create a high resolution LRGB image. The
dots are resolved OK, but there may have been some vibration during the
long exposure and I forgot that DIC slightly reduces resolution in one
direction. So there's room for further improvement. Sigh. There's
always *something* I forget to tweak! ;-)
These are very good and classical specimens for LM but also apply to
SEM. What is clear is that nature is pretty much consistent but not
all that consistent based on looking at AP.
Funny you should mention that. While chasing the ultimate LM resolution
I've taken many images of diatoms. At first I was struck by the perfect
regularity of the striae and pores but then I began noticing occasional
'mistakes' . Discontinuities in the patterns, inclusions, missing and
extra pores, stuff like that. I'm slowly archiving close-ups of
imperfections from various species as I find them. They're not a patch
on your SEM images of course, but as the archive grows it may
eventually reveal something of interest. Clever nano-engineers these
diatoms...
With regard to "what else". Some of the other classic test diatoms like
S. Gemma and P. Angulatum would be ideal - particularly as they're less
challenging (bigger) for those of us dealing with mere photons ;-)
